Choroidal thickness profile SD-OCT analysis in dogs affected by GPRA

Abstract

Purpose: To determine choroidal layers thickness in mixed breed dogs affected by GPRA in comparison with age matched healthy dogs using SD-OCT. Methods: The study was performed on 47 dogs which were divided into two groups: 22 dogs diagnosed with GPRA aged 2-12 years and 25 healthy dogs aged 2-12 years. All dogs were examined using slit- lamp biomicroscopy, tonometry, ophthalmoscopy, fundus camera and SD-OCT (Topcon 3D OCT 2000). Measurements of choroidal layers: RPE-Bruch membrane-choriocapillaris complex (RPE-BmCc) with tapetum lucidum in tapetal fundus, medium-sized vessel layer, (MSVL), large vessel layer with lamina suprachoroidea and (LVLS) whole choroidal thickness (WCT) were taken manually with the use of a caliper function integrated with the OCT software. They were performed dorsally (D), ventrally (V) in the distance 5000-6000 μm temporally (T) and nasally (N), in the distance 4000-7000 μm to the optic disc on enhanced depth scans. The measurements were conducted temporally and nasally both in tapetal(Tt,Nt), and nontapetal (Tn,Nn) fundus. Statistical analysis was performed using Statistica 10 software (Mann Whitney U Test). Results: In all dogs affected by GPRA there was a statistically significant (p<0.005) thinning of MSVL in all measured regions (mean[μm]± SD) – control, GPRA respectively, D (33±6)(25±8), V (25±6)(17±4), Tt (48±10)(32±12), Tn (29±7)(16±4), Nt(34±5)(23±4), Nn (29±8)(19±3). Statistically significant thinning of LVLS and WCT was found in all non-tapetal areas (p<0.005 and p<0.01). Conclusions: GPRA in mixed breed dogs is associated with choroid disorder such as thinning of large vessel layer and whole choroidal thickness in non-tapetal fundus as well as the medium vessel layer in all fundus regions.
