Influence of medium type and culture method on the multiplication efficiency of Sternberg’s herb (Chlorophytum comosum (Thunb.) Jacques)

Abstract

Responding to the challenges posed by modern plant micropropagation techniques, a promising technology of shoot culture temporarily immersed in nutrient medium (temporary immersion system TIS) has been developed using SETIS™ bioreactors. This technology reduces production costs through automation and, by providing optimal growth conditions, leads to maximum regeneration efficiency and high-quality products. In this experiment, the usefulness of this technology for the propagation of the Sternberg’s herb (Chlorophytum comosum (Thunb.) Jacques) – a species belonging to ornamental plants but also valuable for its medicinal properties, was assesed. The extract obtained from its tubers has a potential anti-proliferative effect on four human cell lines: HeLa, CCRF-HSB-2, HL-60, U937. In addition, the methanol extract obtained from the leaves can be a valuable source of numerous bioactive compounds used in the form of dietary supplements, ingredients with biological effects and underestimated therapeutic potential. In vitro culture was carried out using the conventional technique on solidified agar (control) and liquid media using the SETIS™ bioreactor. In addition, in both variants, two culture media differing in macro- and micronutrient content (Murashige & Skoog and Rugini) were evaluated, while having the same set of phytohormones: 1 mg × [dm]^(-3) BAP, 0,1 mg × [dm]^(-3)  IBA, 0,1 mg × [dm]^(-3) GA3.  Explants obtained from the flower stalk of the herb plant were used to establish the culture. The effectiveness of the cultures after 4 and 6 weeks was assessed as the number of regenerated shoots, the length of the shoots and their fresh weight. The experiment made it possible to conclude that the highest values of all analyzed parameters were found for bioreactor culture on Rugini medium. The multiplication of shoots in bioreactor culture on Murashige & Skoog and Rugini medium after a period of 4 as well as 6 weeks was higher in comparison with the conventional method. Also, the fresh weight of the obtained shoots and their length (average) in the studied periods was higher on the liquid medium. The results indicate that bioreactor culture using TIS technology allows for higher results compared to the conventional method. The conducted research can be considered as a first step towards the production of Chlorophytum comosum (Thunb.) Jacques on a larger scale.
