Analysis of the phenotypic diversity of the Enterobacterales microflora of the digestive system as a physiological biomarker of land snails of the genus Cepaea from Polesie

Abstract

Land snails are an important biological bioindicator of the state of the environment. The direct impact of anthropogenic factors and the state of the ecosystem on the functioning of these animals may be reflected in the profile of microorganisms associated with their digestive system. The research hypothesis assumed that an environmental stressor of anthropogenic origin causes internal fluctuations in the studied molluscs. The aim of the study was to determine the impact of an exogenous environmental stressor on the phenotypic diversity of the Enterobacterales microflora of the digestive system of land snails Cepaea sp. from Polesie. The research material included land snails of the genus Cepaea, which were collected at a site under the influence of anthropogenic factors and at a site without its influence. Feces obtained from land snails were placed in NB multiplication broth (Biomaxima). After incubation, the material was transferred to differentiation media – chromID™ ESBL (bioMérieux) and SS (Biomaxima) media. Selected bacterial isolates were identified using API 20E tests (bioMérieux). The drug susceptibility of isolates to 12 chemotherapeutics (Biomaxima) was tested using the disc diffusion method on Mueller-Hinton agar (Biomaxima), i.e. CTX 5, CIP 5, CN 10, TOB 10, TPZ 36, SXT 25, IMP 10, C 30, S 10, TE 30, TS 30 and AM 10. The results were interpreted according to EUCAST v. 11.0 of 2021, Societe Francaise de Microbiologie Recommandations 2013. Enterobacter cloacae was isolated from land snails originating from a site under the influence of an anthropogenic factor and without its influence. No bacterial growth was observed on chromID™ ESBL. The tests showed differences only in the case of sodium thiosulfate, indole and gelatin (API 20E tests). Some isolates showed resistance to chemotherapeutics, such as ampicillin, tetracycline and imipenem. The obtained results indicate that the studied land snails of the genus Cepaea from Polesie are vectors carrying potentially epidemiologically important Enterobacterales bacteria (Enterobacter cloacae).
