Preparation of potentiometric sensors for drug analysis containing chlorhexidine digluconate

Abstract
Ion-selective electrodes (ISE-s) belong to the group of potentiometric sensors which analytical signal (voltage) is related to ion exchange reaction or complexation with an appropriate electroactive component. An interesting and also novel field of application is the use of ISE-s to evaluate the effectiveness of masking the bitter taste of many medicinal products using a variety of numerical procedures (the so-called electronic tonque system). This paper presents the results of the study of the construction and parameters of the ISEs with different selectivity destined for testing pharmaceutical samples containing chlorhexidine digluconate. This is a compound with bactericidal and fungicidal activity, used in many oral preparations to which many sweeteners from the group of sugar alcohols, esters and sucrose derivatives are added. In addition, chlorhexidine is also widely used in veterinary medicine to decontaminate wounds and to treat runny and watery eyes. During the research work, ion-selective electrode bodies (Philips IS-561) were used. Three ion-selective electrodes with different membrane composition (% w/w), plasticizers, ion exchangers, were constructed. Chlorhexidine digluconate (20% w/v), and compounds used as excipients in preparations, i.e.: d-sorbitol, xylitol, aspartame, sucralose, acesulfame, propylene glycol were used to prepare the solutions. Measurements of the electromotive force of the system were performed in the following measuring cell: Ag, AgCl | internal solution | membrane | sample solution || 1M CH3COONa solution || reference electrode. Potential measurements were recorded using the EMF Interface System (Lawson Labs. Inc. USA) and PC computer. The calibration functions of the obtained electrodes in solutions of the main ion-chlorhexidine and interfering ions were determined over a concentration range of 10–6 – 5 × 10–3 M. Electrode 1 is characterized by the following analytical parameters: sensitivity 26.1mV/decade, linear range 10–6 – 10–3 M, standard potential E0 = 182.2 mV and correlation coefficient R2 = 0.998. Electrode 2 is characterized by: sensitivity 30.9 mV/decade, linear range 10–6 – 10–3 M, E0 = 276.0 mV and R2 = 0.999. For electrode 3, a sensitivity 41.6 mV/decade, linear range 5 × 10–5 – 5 × 10–3 M, E0 = 70.5 mV, R2 = 0.979 were obtained. All electrodes show a varied response to selected interferents. The developed electrodes can be used to create a sensor array for the chemometric analysis of compound chlorhexidine samples.
