Genetic diversity of Polish Blumeria graminis f. sp avenae population

Abstract
Understanding the genetic structure of Blumeria graminis f. sp avenae is an important aspect of research into combating, monitoring and controlling the presence of this pathogen, which causes a significant loss of oat crops every year. These analyze make it possible to know the evolutionary potential of a population by determining their genetic variation. There are no data in the literature on this issue in relation to Blumeria graminis f. sp avenae. The aim of this study was to estimation of the genetic variability of Blumeria graminis f. sp avenae isolates from different part of Poland by using ISSR (Inter Simple Sequence Repeats)and SCoT(Start Codon Targeted) marker systems. The subject of the research were 40 single-spore isolates collected in different part of Poland in 2020. A binary matrix (0-absence product, 1-presence product) on the basis of the bands patterns obtained as a result of analyses with the use of 14 ISSRand 12 SCoTmarkers was used to determine the frequency of polymorphic bands as well as PIC (Polymorphic Information Content) and RP (Resolving power) values. In addition, this matrix was used to create similarity matrices (SI) according to the Dice’a formula and to construct dendrograms using the UPGMA (Unweighted Pair Group Method with Arithmetic Mean) in the Past program. Both marker systems proved to be useful for the assessment of genetic variability, but the ISSR generated a greater frequency of polymorphic bands and were characterized by higher PIC and RP values. On the basis of the similarity matrix, it was observed that the isolates from Poland showed little genetic diversity. The highest genetic variation was noted for isolates from central Poland, while the smallest for western Poland. Moreover, the clustering of isolates obtained by the UPGMA method was consistent with the geographic location of the isolates.
