The effect of selected growth regulators and biostimulant on acclimation of Echinacea purpurea ‘Raspberry Truffle’

Abstract
Micropropagation allows to quickly obtaining high numbers of good quality and healthy plants. However, during the last stage of the procedure, i.e. acclimation a high mortality rate may occur. This is due to transfer of plantlets from sterile laboratory conditions to the exterior environment where the conditions are stressful for young non adapted plants. Stress during acclimation may be minimized by using growth regulators or biostimulants which improve a general plant condition. The aim of this work was to develop a protocol of acclimation ex vitro of microcuttings of Echinacea purpurea ‘Raspberry Truffle’ – an important ornamental plant. The following preparations/substances were used: water solution of biostimulant Goteo (0.3%) and growth regulators: abscisic acid (ABA 0.001%), indole-3-butyric acid (IBA 0.2%), brassinolide (BR 0.001%) and epibrassinolide (BL 0.001%). The experimental material came from the in vitro cultured plants. Microcuttings were acclimated in plastic boxes filled with the mixture of peat and perlite (2:1, v/v), pH 5.0-6.0, placed in a phytotrone at 25oC under 16 h day (50 μmol·m-2·s-1). There were 6 treatments, each containing 10 microcuttings. After 6 weeks in phytotrone, plants were transferred to the greenhouse. Results shows significant effect of brassinolide, epibrassinolide and biostimulnat on growth parameters of young plants of analyzed species. Brassinolide affected the highest survival rate of plants (90%). It was observed the effect of biostimulant on length of roots and leaves. The highest plants were obtained by using biostimulant. Brassinosteroids, abscisic acid and biostimulants increased the chlorophyll content (SPAD) in leaves. Treatment on preparations/substances affects on stomatal conductance in leaves as well.
